World number three Tsitsipas had previously defeated Christian Garin in straight sets in the third round in Miami, and before that went without a fight as rival Richard Gasquet pulled out of the match.
Tsitsipas started the year promisingly, winning his first ten matches before losing to eventual champion Novak Djokovic in the Australian Open final. He came back to winning ways in the Davis Cup, winning both singles matches against Ecuador before reaching the round of 16 in Rotterdam.
The 2021 quarter-finalist is currently 8-4 in Miami, having reached the fourth round last year (lost to eventual champion Carlos Alcaraz) and also in 2019 (lost to Denis Shapovalov).
Meanwhile, Khachanov is 11-5 in Miami in 2023 with a pair of wins this week over Thomas Martin Etcheverry and Jiri Legechka.
Earlier this season, Khachanov reached the quarter-finals of Adelaide 1 before finishing in the top four at the Australian Open.
Khachanov is 4-6 in Miami.
Tsitsipas has an excellent 6-0 record against Khachanov, dropping just three sets against him, including one in their last meeting in the Australian Open semi-finals this year. All but one of their encounters have been on hard courts.
